dc.description.abstract | The use of natural products as a way to reduce the use of chemicals in production has been
gaining prominence in the means of animal production. The present study aimed to analyze
the antimicrobial activity of Terminalia catappa dry leaf extracts in vitro on the antimicrobial
growth capacity of Aeromonas hydrophila and Staphylococcus aureus. The strains of
Aeromonas hydrophila and Staphylococcus aureus used were isolated from naturally infected
fish with lesions compatible with aeromonose from commercial fish culture. The bacteria
were reactivated by overnight cultivation in TSB broth and then cultivated on soybean
tryptone agar for 18 hours. suspended in 0.9% NaCl saline to the MacFarland 0.5
concentration. To obtain the extract, 60 grams of the ground material wrapped in a filter paper
(Whatman) was weighed and hot extracted in the Soxhlet extractor apparatus with the aid of
the PA alcohol solvent, then the extract was evaporated in a rotary evaporator and stored in
vials. Four concentrations of 100mg of sample for 2.5 and 5 ml of distilled water and 200mg
of sample for 2.5 and 5 ml of distilled water were used for in vitro tests. Petri dishes (150x20
mm) containing 60 ml were used. of Mueller Hinton agar (KASVI), being prepared
and inoculated with the suspension of Aeromonas hydrophila and Staphylococcus aureus with
the aid of swab. After that, 6 mm paper discs were inoculated with 10 µl of the tested extracts
and placed on the previously prepared plate, where discs without inoculation of extracts were
also added. The assay was done in 3 repetitions each. The plates were incubated at 37 ° C for
24 hours and the formation of inhibition halos around the discs was evaluated. The results
showed that the concentrations of Terminalia catappa extract (100 mg sample for 2.5 and 5
ml distilled water and 200 mg sample for 2.5 and 5 ml distilled water) showed no inhibition

dc.description.resumo | O uso de produtos naturais como forma de diminuir o uso de químicos na produção vem
ganhando destaque nos meios de produção animal. O presente estudo teve como objetivo
analisar a atividade antimicrobiana dos extratos das folhas secas da Terminalia catappa in
vitro sobre a capacidade antimicrobiana no crescimento da Aeromonas hydrophila e
Staphylococcus aureus. As cepas de Aeromonas hydrophila e Staphylococcus aureus
utilizadas foram isoladas de peixes naturalmente infectados, com lesões compatíveis com
aeromonose, proveniente de piscicultura comercial, as bactérias foram reativadas por cultivo
overnight em caldo TSB e então cultivada em meio ágar triptona de soja por 18 horas e
suspendidas em salina NaCl 0,9% até a concentração 0,5 da escala MacFarland. Para
obtenção do extrato foi pesada 60 gramas do material moído envolvido em um papel filtro
(Whatman) e feito uma extração a quente no aparelho extrator de Soxhlet com o auxílio do
solvente a base de álcool etílico PA, em seguida o extrato foi evaporado em um evaporador
rotativo e armazenado em frascos. Foi utilizada quatro concentrações de 100mg de amostra
para 2,5 e 5 ml de água destilada e 200mg de amostra para 2,5 e 5 ml de água destilada, para
os testes in vitro foi utilizado placas de petri (150x20 mm) contendo 60 ml de ágar Mueller
Hinton (KASVI), sendo preparadas e inoculadas com a suspensão de Aeromonas hydrophila
e Staphylococcus aureus com o auxílio de swab. Após isso, discos de papel de 6 mm foram
inoculados com 10 µl dos extratos testados e colocados sobre a placa previamente
confeccionada, onde também foram adicionados discos sem inoculação de extratos sendo esse
o controle negativo e o antibiótico clororanfenicol (30µg) utilizado como controle
positivo. O ensaio foi feito em 3 repetições cada. As placas foram incubadas a 37 ºC por 24
horas e posteriormente avaliou-se a formação de halos de inibição ao redor dos discos. Os
resultados demonstraram que as concentrações do extrato da Terminalia catappa (100 mg de
amostra para 2,5 e 5 ml de água destilada e 200 mg de amostra para 2,5 e 5 ml de água
destilada) não apresentaram a formação do halo de inibição. | pt_BR |
