Efeito de diferentes balanços de reguladores de crescimento no cultivo in vitro de Manihot esculenta Crantz variedade BRS Kiriris

The Manihot esculenta Crantz species has wide global and national economic relevance for its
important value in human, animal and industry. The vegetative propagation of this species
manifests some obstacles as diseases, and micropropagation appears as a viable alternative to
traditional methods of propagation. The objective of this research was to evaluate the action
of different concentrations of the growth regulators 6-benzylaminopurine (BAP),
naphthaleneacetic acid (ANA) and gibberellic acid (AG3) in the in vitro development of
Manihot esculenta Crantz variety BRS Kiriris. The material used as explant source were
Kiriris caulin apices. Explants were introduced into flasks containing 5 ml of the Murashige
and Skoog culture medium supplemented with different concentrations of the regulators. Four
culture media were evaluated: control without growth regulators, constituting treatment 1;
0.01 mg / L of naphthaleneacetic acid - ANA + 0.02 mg / L of benzylaminopurine - BAP +
0.025 mg / L of gibberellic acid - AG3, constituting treatment 2; 0.02 mg / L of ANA + 0.04
mg / L of BAP + 0.05 mg / L of AG3, constituting the treatment 3 (corresponding to the
medium used in the protocol of EMBRAPA - SOUZA et al., 2013) and 0 , 4 mg / L of ANA +
0.08 mg / L of BAP + 0.10 mg / L of AG3 constituting the treatment 4. The experimental
design was a completely randomized (DIC) with 4 treatments, containing 5 replicates, each
replicate consisting of 5 tubes, totaling 25 explants per treatment and 100 explants for the
experiment, were evaluated after 30 days of in vitro cultivation the variables seedling height
(cm), number of green leaves, number of dead leaves, number of roots and root length (cm).
Among the treatments, the best treatment for the variables was treatment 2 (0.01 mg / L ANA,
0.02 mg / L BAP and 0.025 mg / L AG3). Therefore, the addition of growth regulators to the
culture medium is promising for the in vitro establishment of the BRS Kiriris variety. The
combination of 0.01 mg / L ANA + 0.02 mg / L BAP + 0.025 mg / L AG3 presented the best
results for the in vitro establishment of the BRI Kiriris variety.
